Block connections from LAN to WAN allow only from LAN to VPN.

Is it possible to block all connections LAN to WAN and only allow from LAN to VPN for certain LAN-ip-clients?

Yes, that is known as policy based routing. Step 4 of this guide: https://docs.opnsense.org/manual/how-tos/multiwan.html
